Well, the ESP would be Japanese and cost more than the USA Jackson. (Unless ESP still has their California custom shop and actually builds there, but it will still cost more than the Jackson).
I feel my soul go cold... only the dead are smiling.

A custom shop guitar, or a guitar built in a low volume factory in Japan or the US will be better than a mass produced guitar coming out of Japan or the US......
The main difference will indeed be the price.
Since ESP is a Japanese-original company (unlike Jackson, Gibson, Fender, etc) there is no pressure from any US office to only make lesser-quality product available to the USA market so there is no direct competition with the USA-made product, like you find with Jackson/Fender/Gibson/etc.
The quality difference between the Japanese-market-only models like the Grover Jackson or Jackson Stars lines and the USA Select line is nearly indistinguishable - I've got a JacksonStars Archtop Soloist that is just as good in build quality as the 1990-96 USA models I have, and the fairly recent Grover Jackson Roswell I have is slightly better than my Professional series Roswell, although it has the pointy headstock and dot inlays. The neck itself feels better (though slightly thicker) and the body is identical. Playability is far superior to my 2001 DK-2 (at least when comparing it to before I swapped my DK-2's original neck).
However, for what my Archtop Soloist cost me to get it from Japan in a TKL hard case, I could have bought a used USA Jackson Soloist with original Jackson form-fit case.
If you're going with a Custom Shop guitar, go with Jackson. It will be cheaper at final delivery because the inport taxes and such on the ESP would crank the expense higher.
Additionally, you get the benefit of more work being done by hand than computer, and I'll be damned before I pay more for putting a human out of work

The most human thing we can do is comfort the afflicted and afflict the comfortable.
Only you can answer the question of which would be easier to play. Both make top equal quality instruments when it comes to the custom shop.
If you want something that would probably be a little cheaper and with even more custom options, check out http://www.gmwguitars.com
I've already ordered my second guitar from them and they are the absolute best.
guys i dont agree with the neck shape thing. I had an m1 (i think it was, neck through) back in the day, and if i closed my eyes, i couldnt tell a difference.
i'd say the quality is the same.. it would just be what you prefer, feature wise. either way you are getting a good guitar, you will just hear brand bashers no matter what you get. trust yourself, and the way it feels.
